发育期锌缺乏对小鼠脑金属硫蛋白表达的影响

摘  要:目的观察发育期锌缺乏小鼠其成年时脑金属硫蛋白(metalothionein,MT)的表达情况,探讨发育期锌缺乏影响脑功能及发育的机制。方法成年怀孕ICR小鼠随机分为5组,各组动物在怀孕开始至出生后第20天(P20)进食不同锌水平实验饲料,使5组分别成为:严重缺锌组(severezincdeficiency,SZD,饲料含锌量1mg/kg)、边缘缺锌组(marginalzincdificiency,MZD,饲料含锌量5mg/kg)、适锌组(adequatezincsupply,AZS,饲料含锌量30mg/kg),高锌配对组(highzincpair-fed,HZP,饲料含锌量100mg/kg)及高锌组(highzincsupplementation,HZS,饲料含锌量100mg/kg)。然后各组实验动物均进食普通市售饲料至成年(70日龄,P70)。采用蛋白印迹杂交技术检测各组动物(P70)脑组织MT的表达量。结果各组实验动物成年(P70)脑组织均有MT印迹,脑MT表达量的顺序为SZD<MZD<AZS<HZP<HZS;SZD组动物脑MT的表达量仅为HZS组的19.4%,MZD组动物脑MT的表达量亦只有HZS组的?Objective To examine the brain metallothionein expression of developmental zinc deficiency mice in adult,and to study the mechanisms of decelopmental zinc deficiency on the function and deveopment of brain. Methods Of the first day of pregnancy,maternal ICR strain mice were randomly allocated into 5 groups:SZD (Severe Xinc Deficiency,1mg Zn/kg diet),MZD (Marginal Zinc Deficiency,5mg Zn/kg diet),AZS (Adequate Zinc Supply,30Zn/kg diet),HZP (High Zinc PairFed,100mg/kg diet) and HZS (High Zinc Supplementation,100mg/kg diet).From embryonic day (EO) to postnatal day 20 (P20),maternal mice were fed with experimental diets,then up to adult (70 of day age,P70),all pups in experimental groups were fed with normal diet.The brain metallothionein of experimental mice were examined with Western blotting on P70. Results There were metallothionein immunostainings in brain of all experimental groups.The concentration of brain metallothionein showed:SZD group<MZD group<AZS group<HZP group<HZS group. Conclusion Reduction of adult brain metallothionien expression might result from developmental zinc deficiency and that might be one of the mechanisms of brain function damage.
