摘  要:目的建立DNA原位杂交方法(ISH)检测移植患者外周血白细胞中的人巨细胞病毒(HCMV),并与HCMV-pp65抗原血症(免疫组织化学法,IHC)结果比较。方法选取本院肾移植患者标本60例,移植前后采集外周血标本250例,分离获得白细胞经涂片固定后分别采用ISH和IHC检测HCMV。结果ISH检测阳性率高于IHC,ISH检测的HCMV DNA阳性细胞见于外周血单个核细胞,IHC检测的HCMV抗原阳性细胞主要见于中性粒细胞,ISH检出的阳性细胞数明显高于IHC,且显然早于IHC。结论原位杂交检测HCMV DNA,敏感性高、特异性强,有利于早期、快速监测HCMV的激活。Objective To determine Human cytomegalvirus (HCMV) DNA in peripheral blood leukocytes (PBL) from transplant patients by in situ hybridization(ISH) and compare the result with HCMV-pp65 antigenemia. Metbods 250 blood samples of 60 transplant patients in Ruijin hospital were analyzed by ISH and immunohistochemistry (IHC). Results The positive rate for detection of HCMV DNA by ISH was higher than by IHC. The positive cells detected by ISH and IHC were monocytes and neutrophils respectively and the detection of HCMV DNA was earlier than pp65-antigenemia. Conclusions ISH is useful for monitoring the reactivation of HCMV in early stage after transplantation, it is more specific and sensitive.
