摘 要:目的利用小分子干扰RNA(siRNA)表达质粒转染HTB-94软骨肉瘤细胞,观察其在人体软骨肉瘤细胞HTB-94中对肿瘤细胞生长的影响。方法设计并合成高泳动家族性别决定基因9(Sox9)siRNA,将其转染HTB-94肿瘤细胞,观察肿瘤细胞的Sox9基因的mRNA和蛋白表达、生长曲线和细胞凋亡情况。结果Sox9siRNA的插入片段测序结果与合成的siRNA结果一致,Sox9基因的mRNA和蛋白表达降低,HTB-94肿瘤细胞的生长繁殖受到抑制,细胞凋亡增加。结论Sox9siRNA表达质粒可以通过稳定转染HTB-94肿瘤细胞,抑制Sox9基因的mRNA和蛋白的表达,抑制肿瘤细胞的生长繁殖,同时使肿瘤细胞凋亡。Objectives To construct small interfering (siRNA)Sox9 expression plasmid and transfer it into human chondrosarcoma cells HTB-94, and to check the mRNA and protein expression of Sox9 and cell growth and apoptosis of HTB-94 human chondrosarcoma cells. Methods siRNA (Sox9) expression plasmid was designed and synthesized. And it was transfered into HTB-94 human chondrosarcoma cells. Then the expression of the mRNA and protein of Sox9, cell growth and apoptosis in transferred HTB-94 human chondrosarcoma cells were checked. Results The recombimant plasmid was confirmed by enzyme digestion analysis and DNA sequencing. The expression of the mRNA and protein expression of Sox9 in transferred HTB-94 were significantly reduced. The cell growth of HTB-94 was inhibited, and the apoptosis of HTB-94 was remarkably increased. Conclusion siRNA (Sox9) expression plasmid could be transferred into HTB-94 human chondrosarcoma cells. And it can reduce the mRNA and protein expression of the HTB-94, inhibit the cell growth and cause the apoptosis of the tumor cells.
