摘  要:目的观察补阳还五汤对凝血酶敏感蛋白(Thrombospondin,TSP)1和2及其受体CD36 mRNA在实验性脑出血大鼠脑内表达的影响,以探讨补阳还五汤治疗脑出血的机制。方法将SD大鼠随机分为假手术组、模型组、补阳还五汤组,采用胶原酶诱导建立大鼠脑出血模型,运用逆转录-聚合酶链式反应(reverse transcrip—tion—polymerase chain reaction,RT—PCR)观察脑出血后TSP-1、TSP-2和CD36 mRNA表达变化。结果在脑出血第4天,TSP-1表达达高峰;TSP-2在第14天表达达高峰;CD36在脑出血后第4天、第21天呈双峰表达。补阳还五汤组TSP—1 mRNA表达在第1~4天明显高于模型组(P<0.01);TSP-2 mRNA在第14天达高峰且第1~21天均高于模型组(P<0.05);CD36 mRNA第1~4天表达低于模型组(P<0.05),第14~21天明显高于模型组(P<0.01)。结论补阳还五汤能调节TSP-1、TSP-2和CD36 mRNA在脑出血后的表达而促进新生血管成形、成熟,有益于脑出血后损伤组织的修复。Objective To approach the mechanisms of Buyang Huanwu decoction(BHD)by observing its effects on expressions of thrombospondin - 1 (TSP - 1 ), thrombospondin - 2 (TSP - 2) and their receptor CD36 mRNA in the brain of intracerebral hemorrhagic (ICH) rats. Methods ICH was induced by injecting collagenase type VII stereotaxically into right globus pallidus. Sprague - Dawley rats were randomly divided into three groups : sham - operated group, ICH model group, BHD - treated group. The distribution of TSP - 1, TSP - 2 and CD36 were assayed by reverse transcription - polymerase chain reaction (RT - PCR ). Results TSP - 1 arrived at the peak on the 4th day after ICH ( P 〈 0. 01) ; TSP -2 arrived at the peak on the 14th day while CD36 on the 4th day and 21st day . In BHD -treated group, the expression of TSP - 1 mRNA was notably higher than that of ICH model group from the 1st day to 4th day( P 〈 0. 01) ; TSP - 2 mRNA arrived at the peak on the 14th day and was higher than that of ICH model group from the 1st day to 21st day of ICH( P 〈 0. 05) ; CD36 was lower from the 1st day to 4th day of ICH ( P 〈 0.05) but higher from the 14 th day to 21st day of ICH( P 〈 0.01). Conclusions BHD could regulate the expressions of TSP- 1, TSP- 2 and CD36 mRNA in rats after ICH, which may promote the recovery of injured tissues through the formation and maturation of newborn vessels.
