二黄胶囊对大鼠变态反应性脑脊髓炎淋巴细胞亚群和NK细胞的影响

摘　　要：目的观察二黄胶囊对大鼠实验性变态反应性脑脊髓炎(experimental autoimmune encephalomyelitis,EAE)淋巴细胞亚群和自然杀伤细胞(natural kill,NK)的影响。方法80只雄性Lewis大鼠随机分为正常组,完全弗氏佐剂(CFA)对照组和造模组。造模组用髓鞘碱性蛋白(myelin base protein,MBP)与CFA混合制成抗原,建立EAE大鼠模型,并将其分为模型组、激素组和二黄胶囊组。造模后第7,14,28天,用流式细胞仪检测大鼠外周血淋巴细胞亚群及NK细胞;第14,28天取大鼠脑和脊髓进行病理观察。结果CFA对照组、模型组、激素组和二黄胶囊组大鼠的CD4^+在发病前期已明显降低,急性期CD8^+也明显降低,而CD4^+/CD8^+比值明显升高,各组NK细胞均有明显升高。恢复期模型组CD4^+/CD8^+比值明显低于正常组,而激素组和二黄胶囊组则恢复正常。NK细胞除CFA对照组外,其他组基本也达到正常水平。病理观察发现模型组大鼠大脑和脊髓病变表现为小静脉周围炎性细胞浸润形成袖套样改变,核固缩及软化灶。激素组和二黄胶囊组均有不同程度减轻,病灶区域的炎性浸润细胞数较少,袖套样改变少见。结论二黄胶囊能改善EAE大鼠临床症状和评分,缩短病程,减轻EAE大鼠中枢神经系统炎症反应和髓鞘脱失,对淋巴细胞亚群CD4^+、CD8^+、CD4^+/CD8^+比值及NK细胞有一定的调节作用。故认为二黄胶囊防治EAE可能是通过调节细胞免疫系统来实现的。Objective To study the effects of Erhuang Capsule on subgroups of lymphocyte and natural killer（NK） cells in rats with experimental autoimmune encephalomyelitis（EAE）. Methods 80 SPF - grade male Lewis rats were randomly divided into normal group, complete Freund＇ s adjuvent （CFA） control group and modeling group. Rats in the modeling group were injected antigen containing myelin base protein （MBP） and CFA to induce EAE, and then were divided ran- domly into EAE model group, prednisone group and Erhuang Capsule group. On the 7th, 14th and 28th day, lymphocyte subgroups and NK cells in the peripheral blood of rats were detected by flow cytometer, and the pathological changes of rat brain and spinal cord were harvested on the 14th and 28th day. Results The contents of CD4 ^＋ in all groups were significantly reduced compared with the normal group （ P 〈 0.01 ） before onset, the contents of CD8 ^＋ also were significantly reduced in acute period（ P 〈 0.01）, the value of CD4^＋/CD8^＋ and NK cells significantly enhanced（ P 〈 0.05 or P 〈 0. 01）. In the stage of recovery, the value of CD4^＋/CD8 ^＋ in EAE model group was lower than that in the normal group （ P 〈 0.05 ）, but was restored to the normal level in the prednisone group and the Erhuang Capsule group. NK cells is restored to normal level in other groups except the CFA control group. As for the pathological changes, infiltrated inflammatory cells were found around vein and formed sleeve -like shape, the nucleic pyknosis softened focus were presented in EAE model group. The inflammatory cells foci comparatively decreased, and sleeve - like changes were seldom found in prednisone group and Erhuang Capsule group. Conclusion Erhuang Capsule can ameliorate obviously the clinical symptoms of EAE rats, shorten the course of disease, reduce the inflammatory reaction and demyelination in central nerve system, regulate CD4^＋ , CD8 ^＋ subgroups of lymphocytes and their ratio as well as the NK cells.
