Estradiol Promote Myocardial Angiogenesis in a Rat Model of Acute Myocardial Infarction Through Estrogen Receptors

摘  要:目的探讨雌激素对去卵巢大鼠心肌梗死后心脏雌激素受体表达及血管生成的影响。方法结扎去卵巢Sprague-Dauley(SD)大鼠左冠状动脉制作急性心肌梗死模型,给予不同剂量苯甲酸雌二醇干预治疗。放免法测定心肌梗死前和心肌梗死后4周血清雌二醇水平,免疫组化法测定心肌梗死4周后心肌组织中新生毛细血管的数量,Western blot测定心肌组织中细胞核蛋白雌激素受体α、β的表达。结果去卵巢后大鼠血清雌二醇水平显著降低,不同剂量雌二醇干预后各组血清雌二醇水平升高。去卵巢心肌梗死组新生血管数目显著低于保留卵巢急性心肌梗死组及去卵巢后雌激素干预各组,其中,雌激素替代组新生血管数目最高。各实验组雌激素受体α、β蛋白表达均有差异,雌激素替代组表达最高,去卵巢组表达最低。结论体内雌激素同心肌梗死后血管再生有关,雌激素替代治疗有助于增加去卵巢大鼠心肌梗死模型的心肌血管生成,其机制可能是调节心脏雌激素受体α、β介导来发挥作用。Objective To study the influence of estradiol on myocardial angiogenesis and the expression of estrogen receptors in a rat model of myocardial infarction . Methods Acute myocardial infaetion (AMI) model was established by ligation of left anterior descending coronary in ovarieetomized SD rats, which then were treated with different doses of estradiol Benzoate. Estradiol concentration in serum was measured by Radioimmunoassay before AMI and 4 weeks later, mierovaseular density in myoeardium was evaluated with detection of expression of factor by Immunohistoehemistry, and estrogen receptor α and β protein in the myoeardium were detected with Western blot at 28 d after AML Results Estradiol levels in serum significantly degraded after ovarieetomized, but increased after the treatment of different doses of estradiol. Mierovaseular density in myoeardium 4 weeks after AMI in ovarieetomized group was obviously lower than that of acute myocardial infarction group and estrogen therapy groups. Estrogen replacement therapy group had the highest mierovaseular density. The expression of estrogen receptor α and β protein in the myocardium showed significant different, which was the highest in estrogen replacement therapy group and the lowest in ovariectomized group. Conclusion Estrogen improves myocardial angiogenesis in rats after AMI, this effect may be related to the expression of estrogen receptor α and β in myocardium.
