摘 要:目的 评价地氟烷对大鼠脑缺血再灌注时线粒体呼吸链复合体活性的影响,以探讨其脑保护作用的机制。方法 雄性Wistar大鼠40只,体重250~300g,随机分为4组(n=10):假手术组(S组)、缺血再灌注组(I/R组)、1.0MAC地氟烷组(D1.0组)和1.5MAC地氟烷组(D1.5组)。采用前脑缺血再灌注损伤模型,D1.0组和D1.5组缺血前分别吸入1.0MAC和1.5MAC地氟烷40min。再灌注4h时,迅速断头取前脑,密度梯度离心,分离线粒体,分光光度计法测定线粒体呼吸链复合体Ⅰ+Ⅲ、Ⅱ+Ⅲ和Ⅳ的活性,透射电子显微镜观察线粒体病理学结果。结果 与S组比较,I/R组复合体Ⅰ+Ⅲ和Ⅳ的活性降低(P〈0.05);与I/R组比较,D1.0组和D1.5组复合体Ⅰ+Ⅲ和Ⅳ的活性升高(P〈0.05);各组复合体Ⅱ+Ⅲ、D1.0组及D1.5组复合体Ⅲ+Ⅲ和Ⅳ的活性差异均无统计学意义(P〉0.05)。前脑缺血再灌注后,线粒体体积增大、基质电子密度降低、嵴间隙增宽、破裂、膜崩解;D1.0组和D1.5组线粒体超微结构改变不明显。结论地氟烷可减轻大鼠脑缺血再灌注损伤,机制与提高线粒体呼吸链复合体Ⅰ+Ⅲ和Ⅳ的活性有关。Objective To investigate the effects of different concentrations of desflurane on the activities of mitochondrial respiratory chain complexes in a rat model of forebrain ischemia-reperfusion and the possible mechanism. Methods Forty healthy male Wistar rats weighing 250-300 g were randomly divided into 4 groups ( n = 10 each): group Ⅰ sham operation (S); group Ⅱ I/R; group m 1 0 MAC desflurane (D1.0) and group Ⅳ 1.5 MAC desflurane (D1.5 ). The animals were anesthetized with halothane, intubated and mechanically ventilated. PaCO2 was maintained at 35-45 mm Hg. Forebrain ischemia was induced by clamping the bilateral common carotid arteries for 10 min combined with controlled hypotension induced by exsanguination ( MAP was maintained at 43-47 mm Hg) In group D1.0 and D1.5 the animals inhaled 1.0 and 1.5 MAC desflurane for 40 min before ischemia respectively. The animals were decapitated at 4 h after reperfusion was started. The forebrain was immediately removed and the mitochondria were isolated and enriched by homogenization followed by density gradient centrifugation. The activities of mitochondrial respiratory chain complexes including complex Ⅰ + Ⅲ , complexes Ⅱ + Ⅲ and complex Ⅳ were measured by spectrophotometry and mitochondria were examined with electron microscope. Results The activities of mitochondrial respiratory chain complexes Ⅰ + Ⅲ and Ⅳ were significantly decreased in I/R group. ( Ⅱ ) as compared with sham operation group ( Ⅰ ) (P 〈 0.05). Desflurane pretreatment enhanced the activities of complexes Ⅰ + Ⅲ and IV inhibited by I/R in a concentration-dependent manner in group D1.0 ( Ⅲ ) and D1.5 ( Ⅳ ) as compared with group I/R ( Ⅱ ). There was no significant difference in the activities of complexes Ⅱ +Ⅲ among the 4 groups.
