摘  要:采用ISSR—PCR技术为分子标记,对不同地区金钱豹的分子遗传多态性进行研究.采用非损伤性取样方法采集血样或从死亡的样本中取内脏组织,提取基因组DNA,筛选合适的ISSR引物进行随机扩增,用PopGen、Ntsys等软件进行数据分析。检测其遗传分化程度,Nei’s指数计算的遗传一致度和遗传距离以及聚类关系显示,金钱豹9个个体遗传一致度为0.5175~0.8860,遗传距离为0.1211~0.5931,金钱豹不同个体之间存在一定程度的差异,其中宁武的两兄妹个体关系密切,个体之间的遗传关系同性别和地理位置没有明显的对应关系。The genetic diversity of Panthera pardus from 8 different locations was examined using inter- simple sequence repeat (ISSR) markers. The samples of blood or tissue were collected using undamaged sampling methods,then the total genome DNA were isolated and ISSR-PCR finished using chosen primers. The "0" and "1" data were analyzed using Popgene software and the genetic distances were calculated and the phylogenetic tree constructed by using UPGMA method. The results showed that genetic distances between different individuals of P. pardus were between 0. 517 5 and 0. 886 0, showing the genetic differences of P. pardus individuals. The UPGMA tree showed that the two individuals from Ningwu first clustered closely and there are no relative relationship between geographic distance and genetic diversity.
