摘  要:目的:探讨心内直视手术体外循环对犬心肌细胞凋亡的影响及建立稳定的试验犬体外循环模型的方法。方法:6只试验犬采用不同的主动脉阻断时间和辅助循环时间建立体外循环模型,分别于阻断循环前,开放循环前及结束试验前留取心肌标本,采用免疫组化方法检测磷酸化蛋白激酶B(AKt)的表达,TUNEL法检测心肌细胞凋亡指数。结果:开放循环后犬心肌细胞磷酸化AKt表达增多,凋亡指数增加。结论:体外循环术后犬心肌细胞凋亡增多,可能是心脏手术后影响心功能恢复的重要原因,磷酸化AKT的上调尚不足以抑制凋亡的发生。Obiective: To establish the methodology of cardiopulmonary bypass (CPB) experimental model of dogs and to investigate the effects of CPB on myocardial apoptosis in dogs. Methods: Six dogs were underwent CPB with different aortic cross clamping and CPB time. Myocardium samples were obtained just before the aortic cross clamp ,just before the aortic declamping and at the end of the reperfusion. The levels of phosphorated AKt (P-AKt) were detected by immunohistochemistry. Myocardium apoptotic index was detected by TUNEL. Results: The expression of P-AKt in myoeardium and the myocardial apoptotic index were increased significantly after aortic declamping. Conclusion: The increase of myocardium apoptosis after CPB may contribute to the cardiac disfunction after cardiac surgery. The upexpression of p-AKt was not enough to attenuate the apoptosis.
