摘  要:目的:探讨细胞间黏附分子-1(intercellular adhesion molecule-1,ICAM-1)及中性粒细胞(poly- morphonuclear neutrophils,PMNs)浸润在慢性病毒性心肌炎过程中的作用及甘草酸二铵(甘利欣,diammonni glycyrrhizinatis,DG)对其的影响。方法:建立慢性病毒性心肌炎小鼠模型,取心肌用逆转录-聚合酶链反应(RT-PCR)法和免疫组织化学方法检测ICAM-1 mRNA及其蛋白表达水平,用酶法测定PMNs数,酶联免疫方法测定血浆白细胞介素-6(IL-6)的含量。结果:慢性病毒性心肌炎ICAM-1表达水平升高,PMNs增加,血浆IL-6水平升高;甘草酸二铵可抑制慢性病毒性心肌炎心肌ICAM-1的表达、PMNs的增加及血浆IL-6水平的升高。结论:ICAM-1参与、影响慢性病毒性心肌炎的炎症过程,甘草酸二铵可能通过抑制ICAM-1的表达而产生心肌保护作用。Objective: To investigate the effects of diammonium glycyrrhizinate (ganlixin) on the expression of intercellular adhesion molecule-1 (ICAM-1) and myocardial infiltration of polymorphonuclear neutrophils (PMNs) in chronic stage of viral myocarditis (VMC). Methods: After VMC was established in mice, the expression of ICAM-1 mRNA and protein was detected by semi-quantitative RT-PCR and immunocytochemistry. The numbers of PMNs were determined by measuring MPO enzyme activity. The interleukin-6 level in plasma was detected by ElISA method. The results from mice with and without ganlixin were compared. Results: The expression of myocardial ICAM-1 mRNA, the levels of plasma ICAM-1 protein and interleukin-6, and the number of PMNs were increased in VMC mice. Ganlixin significantly attenuated the increased responses in VMC mice (P 〈 0.05 or P 〈 0.01 ). Conclusion: ICAM-1 inducing adhesion and infiltration of PMNs is implicated in the development of chronic stage of VMC. Ganlixin may exert a protective effect on chronic VMC via blocking ICAM-1 expression.
