摘 要:目的:筛选石斛假鳞茎中产生物碱的内生菌并通过培养优化提高次生代谢产物的含量。方法:从新鲜石斛中分离生物碱产生菌,并对其发酵条件进行研究。结果:筛选到一株产生物碱的内生细菌,属于蜡状芽胞杆菌。初步确定了最适的发酵培养基组成为:去皮土豆20%,葡萄糖2%,蛋白胨0.25%,硫酸铵0.25%,磷酸二氢钾0.1%,硫酸镁0.05%,氯化锰0.01%,pH值6.5;最适的发酵条件为:接种量6%~8%,温度28~30℃,培养时间48h。结论:通过初步优化,单位体积发酵液的生物碱含量较原始条件大幅提高。Objective:To select the endophytes in Dendrobium nobile Lindl and increase the yield of alkaloids by improving fermentative conditions. Methods: The strain of endophytes which could produce alkaloids was isolated from Dendrobium nobile Lindl, and the fermentative conditions were also studied. Results: A strain of Bacillus cereus was isolated, which probably produce the same secondary metabolite as the hosts do. It was proved that the optimum fermentative conditions for producing alkaloids were the improved potato medium which contained 2% glucose, 0.25% tryptone, 0.25% ammonium sulfate, 0.1% potassium dihydrogen phosphate, 0.05% magnesium sulfate, and 0.01% manganese chloride at p14 6.5 ; inoculation with 12 h' seed-age and 6% - 8% of inocula; and culturing for 48 h at 28 - 30 ℃. Conclusion: With the optimized culture conditions, the yield of alkaloids increases obviously compared with the original cultural condition.
