补阳还五汤对动脉粥样硬化模型主动脉基质金属蛋白酶表达量的影响

摘　　要：目的观察补阳还五汤对动脉粥样硬化大鼠模型的治疗作用,并从蛋白水平探讨其作用机制。方法大鼠动脉粥样硬化造模成功后随机分为补阳还五汤治疗组、川芎嗪治疗组、模型组,另设正常对照组。予相应处理后检测腹主动脉基质金属蛋白酶(MMP-9)表达水平。用药前后取尾静脉血2ml测血清总胆固醇、三酰甘油、高密度脂蛋白胆固醇、低密度脂蛋白胆固醇水平。结果高脂饮食可诱导大鼠形成动脉粥样硬化模型,其主动脉组织MMP-9强阳性表达。补阳还五汤治疗组的MMP-9表达量水平、低密度脂蛋白胆固醇显著低于川芎嗪治疗组、模型组。结论益气活血代表方剂补阳还五汤可以下调MMP-9的表达,降低血脂,具有抗动脉粥样硬化作用。降低主动脉组织MMP-9的表达可能是其作用机制之一。Objective：Making use of atherosclerosis（AS）rat model induced by high fat diet,we have studied the effect of Buyanghuanwu Decoction on AS rat model,meanwhile,investigate the mechanism on protein level.Methods：The AS rats were divided into Buyanghuanwu Decoction group,Ligustrazine group,control group and blank group after model successing.The expression level of Matrix Metalloproteinase-9（MMP-9） and blood-lipid were detected.Results：High fat diet could induce atherosclerosis in rat.Compairing with normal control group,the level of TC,TG,LDL of model group was increased.There was high positive expression of MMP-9 in atherosclerosis model aorta.Compairing with ligustrazine therapy group and model group,the expression of MMP-9,the level of LDL of Buyanghuanwu Decoction group were decreased significantly.Conclusion：Representative formula of Qi-Benifiting and Blood-Activating Chinese medicinal herbs Buyanghuanwu Decoction have effect of down-regulating the expression of MMP-9,lowering the level of blood-lipid in rat model,so that have effect of againsting atherosclerosis.Down-regulating the expression of MMP-9 may be one of the mechanisms of againsting atherosclerosis.
