摘 要:目的建立夏枯草中齐墩果酸和熊果酸的含量测定方法。方法以布洛芬为内标,采用毛细管区带电泳,高频电导检测;未涂层弹性融硅石英毛细管柱55cm×75μm ID,有效长度46cm;1.2mmol/L三乙胺-HCl(pH=10.60),β-环糊精(0.08mmol/L)为运行缓冲液;分离电压12.5kV;重力进样10s(高度20cm)。结果齐墩果酸、熊果酸线性范围分别为4.1-114.8μg/mL(r=0.9995)和4.3-120.4μg/mL(r=0.9996);平均回收率分别为96.7%和98.2%。结论该法简便、准确、快速、重现性好,可用于夏枯草药材中齐墩果酸和熊果酸的含量测定。Objective To establish a method for determining the content of oleanolic acid and ursolic acid in Prunella vulgaris. Methods Capillary electrophoresis with high frequency conductivity detector was used to analyze the sample. The separation was performed on a uncoated fused silica capillary of 55 cm × 75 μm(46 cm effective length) using 1.2 mmol · L^-1 triethylamine as washing buffer (containing0.08 mmol· L^-1 β-cyclodextrin, pH 10.60). The voltage was 12.5 kV and temperature was 25 ℃. The sample was injected by gravity( 10 s,20 era). Results The linear ranges of oleanolic acid and ursolic acid were 4.1- 114.8 μg/mL( r = 0.9995 ) and 4.3 - 120. 4 μg/mL ( r = 0. 9996) , respectively. The average recoveries of oleanolic acid and ursolic acid were 96.7% and 98. 2%, respectively. Conclusion This method is reliable, rapid and accurate, which could be used for the quantitative analysis of the contents of oleanolic acid and ursolic acid in Prunella vulgaris.
