摘 要:目的:探讨大鼠胃组织和血浆ghrelin表达水平对普通饲料诱导的肥胖倾向(obesity-prone,OP)与肥胖抵抗倾向(obesity-resistance-prone,ORP)形成的作用。方法:16只健康雄性Wistar大鼠,普通饲料(脂肪供能比为14.8%)喂养10周后,根据其腹脂/体重比值大小选入OP组与ORP组,每组7只。记录体重和摄食量,测定内脏脂肪含量和空腹血糖、血脂、血胰岛素和血浆ghrelin水平,以及胃组织ghrelin蛋白表达水平。结果:OP组大鼠体重增量、能量利用率、内脏脂肪含量明显高于ORP组(P<0.01);与ORP组大鼠相比,OP组血糖、甘油三酯、胰岛素水平显著升高(P<0.05),血浆ghrelin水平有降低趋势(P=0.14);OP组大鼠胃组织ghrelin表达显著高于ORP组(P<0.01);大鼠胃组织ghrelin表达水平与体重增量、能量利用率、肾周与睾周脂肪、空腹血糖呈正相关关系(P<0.05),与胰岛素敏感指数呈负相关关系(P<0.05)。结论:胃组织ghrelin表达水平较高或较低很可能是大鼠形成OP或OPR的重要影响因素。Objective To explore whether ghrelin in stomach and plasma play roles on development of obesity- prone( OP) and obesity-resistance-prone(ORP)rats induced by a regular chow. Methods Sixteen male Wistar rats were fed with a regular chow with 14.8% of energy from fat for 10 weeks. Body weight, food intake, and visceral fat were measured. The rats were divided into OP or OPR group according to their visceral fat weight/body weight. There were 7 rats in each group. Glucose,TG, TC, HDL-C, LDL-C, insulin and ghrelin in fasting plasma were assayed. Ghrelin expression in stomach was investigated by Western Blot. Results Body weight gain,energy efficiency,and visceral fat depots in OP rats were significantly higher than those in ORP rats(P〈0.01);Fasting plasma glucose,TG,insulin levels in OP rats were increased compared to ORP rats (P〈0.05);When compared with ORP rats, OP rats had an increased tend in plasma ghrelin level(P= 0. 14). OP rats had significantly elevated ghrelin expression in stomach than did ORP rats(P〈0.01). Stomach ghrelin expression of rats was positively correlated(P〈0.05) to body weight gain, energy efficiency, weights of perirenal and epididymal fat pads, fasting plasma glucose, but negatively correlated(P〈0.05)to insulin sensitive index(ISI). Conclusion The higher or lower ghrelin expression in stomach may be associated with OP or OPR development in the rats.
