摘 要:目的比较不同配比的吗啡和氯诺昔康复合应用于切口痛大鼠的镇痛效果。方法雄性成年SD大鼠63只,依Brennan法建立大鼠切口痛模型,随机分为假手术组(A组):吸入1.4%异氟醚5min后不做处理;切口痛对照组(B组):切口痛模型大鼠,皮下注射1ml生理盐水;给药组(C^H及L组):切口痛模型大鼠,分别给予不同配比的吗啡/氯诺昔康(6/0、1/5、2/4、3/3、4/2、5/1、0/6mg.kg-1),皮下注射。每组7只大鼠,均分别于术后1h给药,然后采用累积疼痛评分法评价各组药物对大鼠的镇痛作用。结果A组的累积疼痛评分为0.375±0.518明显低于其他给药组(P<0.01),B组的累积疼痛评分为18.125±1.126显著高于其他给药组大鼠(P<0.01);C、D、E、H、L组各组间比较均无统计学意义(5.250±1.488、5.375±1.506、5.125±1.458、4.375±2.200、5.250±2.121,P>0.05);F、G组的累积疼痛评分分别为3.625±1.061和3.750±1.165显著低于C、D、E、L组(P<0.05或P<0.01)。结论吗啡与氯诺昔康复合应用的比例在1∶1~1∶0.5时可取得较佳的镇痛效果。Objective To compare the analgesic effect of morphine combined with lornoxicam in different proportions in rats. Methods 63 male adult SD rats were assigned randomly into nine groups (7 each). Under the 1.4% isoflurane anesthesia, an incision of 1cm in length was made in the plantar region of the left hind paw parallel to the muscle according to the method of Brennan. The rats in sham operation group (A) inhaled 1.4% isoflurane for 5 min but no incision was made, the rats in operation control group (B) received hypodermic injection of 1ml 0.9% NaCl 1h after incision, the rats in groups C to H and group L received hypodermic injection of morphine combined with lornoxicam in different proportions (6mg/kg∶0mg/kg; 1mg/kg∶5mg/kg; 2mg/kg∶4mg/kg; 3mg/kg∶3mg/kg; 4mg/kg∶2mg/kg; 5mg/kg∶1mg/kg and 0mg/kg∶6mg/kg, respectively) 1h after incision. Analgesic effect was assessed by cumulative pain score within 1 hour after administration. Results The cumulative pain scores of rats in group A was 0.375±0.518, which was significantly lower than that of the other groups (P〈0.01). The cumulative pain scores of rats in group B (18.125±1.126) was significantly higher than that of the other groups, in which the animals received combined morphine and lornoxicam administration (P〈0.01). The scores of groups C, D, E, H and L did not show statiscally significant differences compared with each other (5.250±1.488, 5.375±1.506, 5.125±1.458, 4.375±2.200 and 5.250±2.121, respectively, P〉0.05). The scores in groups F and G were 3.625±1.061 and 3.750±1.165 respectively, which were significantly lower than those in groups C, D, E and L (P〈0.01 or P〈0.05). Conclusion Morphine combined with lornoxicam in proportion of 1∶1 to 1∶0.5 provides more powerful analgesic effect because of their potential synergistic effect.
