摘 要:选用生物凝胶A为载体,经交联、脱硫、筛选、溴化氰活化后,再共价结合抗重组人肿瘤坏死因子-a(rHuTNF-α)单克隆抗体,观察抗体偶联率及该免疫吸附剂体外吸附清除TNF的效能。结果表明:①随着加入抗体量的增加,载体偶联的抗体量也逐渐增加,但当每毫升凝胶偶联抗体8.12mg时,则基本达偶联量的峰值;②免疫吸附剂能高效清除循环中TNF,而对正常血浆蛋白仅有少许非特异性吸附,且非特异性吸附较易洗脱。There is increasing evidence that tumour necrosis factor α(TNF α) is a principal mediator in the cascade of pathophysiologic events following endotoxic shock. In order to prepare an immunosorbent for removing TNF α from blood, crosslinked and desulfated agarose beads 250 300μm in diameter were selected as matrix particles. Antibodies were then attached to the beads activated by CNBr. The results showed that addition of progressively greater amounts of antibody resulted in attachment of more antibody to the beads. The agarose anti TNF immunosorbent could remove circulating TNF effectively by extracorporeal circulation in vitro, and just a few amounts of plasma proteins were adhered to the beads nonspecifically.
