摘 要:将预培养不同时间的毛泡桐和白花泡桐茎段、叶片和叶柄分别在含有不同浓度秋水仙素的液体、固体和双层培养基上进行四倍体植株诱导,并采用根尖染色体计数和成熟叶片单细胞DNA含量测定进行诱变植株的倍性分析.结果表明,叶片是毛泡桐和白花泡桐四倍体诱导的最适材料,双层培养基处理叶片后四倍体诱导率最高,不经预培养的毛泡桐叶片在质量浓度为2000 mg·L^(-1)的秋水仙素双层培养基上处理72 h和预培养12 d的白花泡桐叶片在质量浓度为1000 mg·L^(-1)的秋水仙素双层培养基上处理24 h分别是毛泡桐和白花泡桐四倍体诱导的最佳组合.Tetraploid induction of Paulownia tomentosa and Paulownia fortunei from their stems, leaves and leafstalks of the diploid with various concentrations of colchicines on liquid, solid, liquid-solid MS media through chromosome numbering of the plant tip cells and DNA content in leaf cell was investigated. The results indicated that leaves and liquid-solid MS medium were the optimal explants and medium for tetraploid induction of the plants respectively. The highest tetraploid induction rates were obtained from the diploidy leaves of P. tomentosa treated on the liquid-solid MS medium with 2 000 mg · L^-1 colchicine for 72 h and the ones of P. fortunei pre-cultured for 12 d first and then treated on the media with 1 000 mg · L^-1 colchicine for 24 h might be induced.
