摘  要:目的:建立双通道高效液相色谱法同时测定人体血清中维生素A、D_3、E的含量,以诊断维生素缺乏症,监视营养状况。方法:取血清100μL,加入100μL乙醇,震荡,加入0.7 mL正己烷萃取,通氮气将正己烷提取液挥发干后用乙醇溶解,进样分析。色谱条件:色谱柱为Nova-Pak C_(18)(3.9 mm×150 mm,4μm),流动相为甲醇-水(95:5),流速为1.5 mL·min^(-1),检测波长维生素A、E为300 nm,维生素D_3为265 nm。结果:维生素A在0.10~0.90 mg·L^(-1)范围内呈线性关系,r=0.999 9,维生素D_3在5.00~45.00 mg·L^(-1)范围内呈线性关系,r=0.999 3,维生素E在1.00~40.00 mg·L^(-1)范围内呈线性关系,r= 0.997 0;维生素A、D_3、E的平均回收率分别为94.7%,90.2%,98.8%。结论:本方法快速、灵敏、准确,简便易行。OBJECTIVE To establish a method to determine the concentrations of retinal, cholecalciferol(D3)andα-tocopherol in human serum simultaneously. METHODS 100 μL serum was added with 100 μL anhydrous alcohol and 0. 7mLhexane,the mixture was shaken and centrifuged, then dried hexane extract with N2 gas. Resolved with anhydrous alcohol. The HPLC system was consisted of Nov-Pak C18 (3.9 mmX 150 mm,4/~m) column, the mobile phase was methanol-water (95 : 5), flow rate was 1.5 mL.min^-1, and the detection wavelength for retinal and a-tocopherol was 300 nm and the detection wavelength for cholecalciferol(I)3 )was 265 rlrru RESULTS The liner ranges of determination of retinal, cholecalciferol(D3 ) andcctocopherol were 0.10 - 0. 90 mg. L^-1(r=0.9999), 5. 00- 45. 00 /μg·L^-l(r =0.9993) and 1. 00 - 40. 00 mg· L -1 (r=0.9970) respec tively. The relative recoveries of retinal, cholecalciferol(D3 )anda tocopherol were 94. 7%, 90. 2%, 98. 8% respectively. CON- CLUSION The method is simple, accurate, sensitive and precision.
