摘 要:目的:探讨NAT2,CYP2B6和GSTP1基因多态性与苯乙烯生物监测之间的关系。方法:选择58名接触苯乙烯的工人为研究对象,应用PCR-RFLP法判定基因型;应用高效液相色谱法测定其尿中三种代谢产物(苯乙醇酸、苯乙醛酸和苯乙烯巯基尿酸)的含量;并分析NAT2,CYP2B6和GSTP1基因多态性对接触不同水平苯乙烯工人尿中代谢产物含量有无显著影响(P<0.05)。结果:CYP2B6(BsrI)不同基因型苯乙烯尿中代谢产物含量有显著性差异;GSTP1(BsmAI)和NAT2 M3基因型在苯乙烯代谢中的作用差异均无统计学意义。结论:除传统生物检测指标如尿中化学代谢物和早期生物效应等,某些苯乙烯代谢酶基因多态性对其生物监测指标具有一定影响,因此也可作为遗传易感性生物标志物,用来评价苯乙烯接触人群的遗传易感性。Objective:To study the relationship between polymophisms NAT2, CYP2B6 and GSTPland biomarkers in styrene -exposed workers. Methods:Polymerase chain reaction -restrained fragment length polymorphism technique (PCR -RFLP) was used to analyze the genetic polymorphisms of NAT2 ,CYP2B6 and GSTP1 among 58 cases of styrene - exposed workers. Results: CYP2B6 mutation genotype was significantly associated with urinary metabolites of styrene, while the genetic polymorphisms of GSTP1, NAT2 M3 were not statistically significant. Conclusion:The present data seem to suggest that apart from the methodology usually used for monitoring populations occupationally exposed to styrene ( urinary metabolites and biomarkers of early biological effects), the analysis of individual genotypes associated with the metabolic fate of styrene should also be carried out to evaluate the individual genetic susceptibility of exposed populations.
