银杏黄酮甙对人巨细胞病毒转染血管内皮细胞的影响

摘　　要：目的观察银杏黄酮甙对人巨细胞病毒(HCMV)感染后血管内皮细胞(VEC)增殖、凋亡及NO分泌量的影响,探讨银杏黄酮对VEC的保护作用。方法应用MTT比色法检测细胞的增殖、流式细胞技术观察细胞凋亡以及硝酸还原酶法检测细胞的NO分泌量。实验分为4组,分别为正常对照组,HCMV感染组,银杏黄酮甙组,HCMV感染+银杏黄酮甙组。结果①HCMV促进人脐静脉内皮细胞(HUVEC304)的增殖(P<0.05)、抑制HUVEC304的凋亡(P<0.01)和降低HUVEC304的NO分泌水平(P<0.01)。②银杏黄酮甙可以促进HUVEC304的增殖并呈剂量依赖性(P<0.05),增加了其凋亡水平(P<0.01),并升高其NO分泌水平(P<0.01)。③银杏黄酮甙可以抑制HCMV感染的HUVEC304的增殖作用(P>0.05),促进其凋亡(P<0.05)并增加其NO的分泌水平(P<0.01)。结论银杏黄酮甙对HCMV感染的HUVEC304具有明显的保护作用,其作用机制可能是抑制HUVEC304增殖并促进NO分泌。Objective To observe the role of ginkgo flavonoid glycoside in human cytomegalovirus （HCMV） infected vascular endothelial ceils （VECs） proliferation, apoptosis and NO secretion and to explore its protective effect on VECs. Methods The proliferation, apoptosis and NO secretion of HCMV-induced human umbilical VEC （HUVEC）304 were measured by MTT assay, flow cytometry method and nitrate reductase method respectively. The cultured HUVEC304 were divided randomly into control, ginkgo flavonoid glycoside, HCMV and ginkgo flavonoid glycoside ＋ HCMV groups. Results （1）HCMV accelerated proliferation （P 〈 0. 05） , inhibited apoptosis level （P 〈 0. 01 ） and decre.ased NO level （P 〈 0. 01 ） of HUVEC304. （2）Ginkgo flavonoid glycoside accelerated proliferation to certain degree in a dose-dependent manner （P 〈0. 05） ,increased apoptosis level （P 〈0. 01 ） and increased NO level （P 〈0. 01 ） of HUVEC304. （3）Ginkgo flavonoid glycoside inhibited proliferation （P 〉0. 05） ,augmented apoptosis （P 〈0. 05） and increased NO level of of HCMV-infected HUVEC304 （P 〈0. 01 ）. Conclusions Ginkgo flavonoid glycoside has obvious protective effects on HCMV-infected VECs, and the mechanism of which may be related to inhibiting the proliferation of HUVEC304 and promoting NO secretion.
