摘 要:目的:探讨苦参碱、γ干扰素(IFN-γ)对博莱霉素诱导的大鼠肺纤维化的干预作用及其可能的机制。方法:SD雄性大鼠60只,分为健康对照组、肺纤维化模型组、苦参碱干预组、IFN-γ干预组,各组分别于7d、14d、28d处死5只,行HE染色及Masson染色半定量判断肺组织肺泡炎及肺纤维化程度。免疫组化测肺组织TGF-β1、CTGF、I型胶原及Ⅲ型胶原的蛋白水平表达。结果:苦参碱干预组、IFN-γ干预组大鼠肺泡炎、肺纤维化程度均较模型组好转。与模型组相比,苦参碱、IFN-γ都能够抑制肺组织中的TGF-β1、CTGF、I型胶原及Ⅲ型胶原的表达(P<0.05)。结论:苦参碱及IFN-γ对大鼠肺纤维化模型均有一定的疗效,其作用机制可能是通过抑制TGF-β1和CTGF的表达来减少Ⅰ型胶原及Ⅲ型胶原的生成。Objective:To evaluate the therapeutic effect and mechanism of matrine and IFN-γin Bleomycin-induced pulmonary fibrosis of rats. Methods: Male SD rats (n = 60) were randomly divided into four groups : (A) normal group, (B) Bleomycin-induced pulmonary fibrosis model group, (C)Matrine group, (D)IFN-γ), group.On days 7,14 and 28 after bleomycin treatment,5 rats in each group were killed, and the lung were harvested for histopathological studies,immunohistochemical determination of TGF-β,CTGF, collagen Ⅰ and collagen Ⅲ. Results:The degree of alveolitis and fibrosis,the level of TGF-β1, CTGF,collagen Ⅰ and collagen Ⅲdecreased significantly in the groups C and groups D, as compared with groups B (P 〈 0.05). Conclusion: The treatment of Matrine or IFN-γhas some protective effects on pulmonary fibrosis in rats ,partly through affecting TGF-β1 and CTGF expression.
