摘  要:目的:建立血浆中多西紫杉醇浓度的高效液相色谱测定方法。方法:以地西泮为内标,血浆样品经叔丁基甲醚3次提取,采用大连依利特C18(250mm×4.6mm,5μm)色谱分析柱;流动相为乙腈-水(47∶53);流速为1.0mL.min-1;检测波长为230nm;进样量为50μL。结果:地西泮和多西紫杉醇的保留时间分别是10.9min和13.2min,多西紫杉醇的最低检测质量浓度为20μg.L-1,血浆样品质量浓度在0.25~16.5mg.L-1范围内与峰面积比线性关系良好,回归方程为C=10.117X+0.428,r=0.9998(n=7),提取回收率为87.24%~92.64%(n=5),方法回收率为97.52%~101.44%,日内RSD为3.6%,2.4%,0.79%,日间RSD为3.8%,2.7%,1.4%,且稳定性良好。结论:本方法简便灵敏,干扰小,可用于多西紫杉醇血药浓度的测定。OBJECTIVE To establish an HPI.C method for determination of docetaxel in human plasma. METHODS Diazepam was used as internal standard. The docetaxel was extracted with tert-bulyl methyl ether three times and determined through ODS-C18 (250 mm× 4. 6 mm, 5 μm)column. The mobile phase consisted of water-acetonitrile (53:47) with the flow rates of 1.0 mL·min^-1. The detection wavelength was 230 nm. RESULTS In this system, the retention times of diazepam and docetaxel were 10.9 min and 13. 2 min, respectively. The detection limit for doeetaxel was 2020μg·L^-1. Docetaxel concentration presented a good linear range of 0. 25-6. 5 mg·L^-1 (r= 0. 999 8). The extraction recoverys were 87. 24%-92. 64% (n = 5) ,and the method recoveries were 97.52%-101.44% ,within-day RSD of 3.6% ,2.4%,0. 79%,and the betweem-day RSD of 3.8%, 2. 7%, 1. 4%. CONCLUSION The method is reliable,convenient and more efficient and can be applied to the determination of docetaxel concentration in plasma.
