摘  要:目的观察谷氨酰胺诱导大鼠肺组织表达热休克蛋白70(HSP70)的作用及HSP70对高氧肺损伤的保护作用。方法健康清洁级雄性Wistar大鼠18只(体重252~286g),随机均分为谷氨酰胺组(G组)和对照组(C组):G组以0.75g/kg谷氨酰胺行腹腔注射,每天注射1次,连续3d;C组大鼠每天腹腔注射同等容积的生理盐水。于注射后第4天每组各取3只大鼠的肺组织,用免疫印迹法(Western blotting)检测HSP70表达,其余12只大鼠全部放入高氧环境(氧浓度95%)中继续喂养,观察在高氧环境下第6天两组大鼠肺组织形态学改变。结果注射谷氨酰胺后第4天,G组大鼠肺组织HSP70蛋白表达水平明显升高,其灰度值为20.34±2.26;C组HSP70蛋白表达水平较低,其灰度值为1.82±0.67,G组明显高于C组(P〈0.05)。G组第6天肺部炎症表现为肺泡内极少量红细胞渗出;而c组病理改变为肺泡大小不等,肺泡腔变大,肺泡壁变薄,有肺大泡形成,肺泡内有出血和炎症细胞浸润。结论大鼠腹腔注射谷氨酰胺可明显增加肺组织HSP70表达,HSP70可减轻高氧肺损伤时肺组织炎性改变。Objective To explore the effects of hyperoxia in the pathogenesis of lung injury and observe the influence of heat shock protein 70 (HSP70) induced by glutamine (Gin). Methods Eighteen Wistar rats were assigned to two groups of Gln group and saline group(C group) with 9 rats each. The G group was treated with Gin 0.75 g/kg via abdomen injection once a day for 3 days and C group was administered with saline. We analyzed pulmonary tissue HSP70 expression by Western blotting on the fourth day. All other rats were fasted in hyperoxia (95%) for 6 days. On the sixth day, the lung histopathology was observed by light microscope. Results Pulmonary HSP70 expression was significantly higher in G group (20.34±2.26) ,than that in C group (1.82±0.67) (P〈0.05) on the 4th day. In G group, very small amounts of red blood cells leaked out into alveoli on the 6th day. In C group, the inequality of size of lung alveoli, hemorrhage and inflammatory cell infiltration in lung alveoli were observed on the 6 th day. Conclusion Glutamine can enhance pulmonary HSP70 expression in the lungs. HSP70 has interventional effect on the pathological process of hyperoxia lung injury.
