摘 要:目的用real-time PCR和RT-PCR检测FP248 mRNA表达,研究其在白血病和肿瘤诊断中的意义。方法用real-time PCR和RT-PCR检测肿瘤细胞株(SGC-7901、A549、95D、SW480、U937、PC3)和白血病(ALL、CML、AML)患者骨髓或外周血以及胃癌等消化道肿瘤患者癌组织中FP248 mRNA的表达水平;用RT-PCR技术检测消化道肿瘤患者外周血单个核细胞(PB-MC)FP248 mRNA的表达。结果SGC-7901细胞株FP248 mRNA强阳性,A549和95D细胞弱阳性,其他细胞株阴性;ALL、CML、AML患者骨髓及PBMC FP248表达上调;胃癌、直肠癌、结肠癌患者癌组织中FP248 mRNA的表达高于癌旁组织,食道癌组织为阴性;胃癌、结肠癌和直肠癌患者PBMC未检测到FP248 mRNA。结论FP 248的表达可能在白血病和消化道肿瘤的发生发展中起重要作用;检测该基因的表达可能有助于白血病和消化道肿瘤的诊断。Objective To investigate the significance of detection for FP248 mRNA in leukemia and solid tumors. Methods RT-PCR and real-time PCR were used to analyze the expression level of FP248 mRNA in the tumor cell lines, i. e. , SGC, A549, D95, SW480, U937, PC3, the bone marrow, and peripheral blood from the patients with leukemia, i. e. , ALL, CML, AML, and the cancer tissues in alimentary tract. FP 248 mRNA in peripheral blood mononuclear cells (PBMC) of the patients with alimentary tract cancer was also detected by RT-PCR. Results FP248 mRNA was strongly expressed in SGC but weakly expressed in A549 and D95 cells. , No expression was detected in the other tumor cell lines. The expression of FP248 mRNA was up-regulated in the bone marrow and peripheral blood of the patients with ALL, AML, CML and the tissues of stomach cancer, colon cancer and rectal cancer comparing with control groups. No FP248 mRNA was detected in esophageal cancer tissue and PBMC of the patients with alimentary tract cancer. Conclusions FP248 gene might play an important role in nosogenesis and development of ALL, CML, AML, stomach, colon and rectal cancer, so it should be a useful marker for diagnosis of leukemia and alimentary tumors.
