云芝糖肽、丹参酮ⅡA对荷瘤小鼠的抗肿瘤及免疫调节作用  被引量:14

摘  要:目的:研究云芝、丹参有效组分单体以及复方对EAC荷瘤小鼠的抗肿瘤和免疫调节作用,为肿瘤的中药治疗新方剂的临床广泛应用提供一定的实验基础。方法:建立EAC实体型荷瘤小鼠动物模型,随机分为6组:生理盐水对照组(NS)、阿霉素治疗组(ADM)、云芝糖肽组(PSP)、丹参酮Ⅱ组(TanⅡA)、复方组(PSP+TanⅡA)和综合治疗组(PSP+TanⅡA+ADM),并给予相应药物治疗,应用免疫组化法检测小鼠脾脏IL-2、IL-2R和肿瘤组织中周期蛋白CDK4以及凋亡蛋白Bax、Bcl-2表达,并计算抑瘤率、脏器指数,结果与对照组和化疗组进行比较。结果:云芝糖肽组小鼠脾脏IL-2、IL-2R水平高于NS组和化疗组(P<0.05),各药物治疗组肿瘤Bax均高于化疗组,CDK4均低于NS组(P<0.05),复方组Bax高于NS组(P<0.05),云芝组糖肽、丹参酮ⅡA组、综合治疗组CDK4低于化疗组(P<0.05),云芝糖肽组、复方组与综合治疗组肿瘤Bcl-2低于NS组(P<0.05),各药物治疗组对肿瘤有明显的抑制作用(P<0.05),复方组胸腺指数高于NS组,各药物治疗组胸腺指数均高于化疗组(P<0.05)。结论:云芝糖肽、丹参酮ⅡA对EAC荷瘤小鼠有明显的抗肿瘤和免疫调节作用,并能缓解化疗药物所引起的毒副作用,二者联用存在一定协同作用。Objective:To study the anti-tumor effects and immtmoregulatory activity of effective ingredients in Yunzhi (PSP) and Danshen (TanⅡ A) by a model of EAC tumor-beating mice,and to provide the fundament to tumor therapy with the traditional Chinese medicines alone and in combination. Methods: Mice were inoculated with EAC ceils and randomized into 6 groups: the control group, ADM group,PSP group,Tan Ⅱ A group,PSP + TanⅡ A group and PSP + TanⅡ A + ADM group. PSP and TanⅡ A were used in alone or in combination. Immunohistochemical methods were employed for detection of IL-2, IL-2R in spleens and CDK4, Bax/Bcl-2 in the tumor tissues of tumor-beating mice. The inhibition rates of tumor growth and thymus index of the therapy groups with the traditional Chinese medicines were calculated and compared with control and ADM groups. Results:The level of IL-2 and IL-2R expressed in spleens was obviously higher in PSP than in both control and ADM group( P 〈 0.05). Expressing level of Bax in tumors, particularly in the PSP + Tan Ⅱ A group, was obviously increased in all the therapy groups than in ADM group ( P 〈 0.05 ) and the control group. CDK4 expression in the tmnor was obviously decreased in the thempy groups vs the control group( P 〈 0.05 ), especially in PSP group, Tan Ⅱ A group and PSP + Tan Ⅱ A + ADM group in which CDK4 expression was lower than in ADM group( P 〈 0.05 ). Bcl-2 expression in the tumor was obviously decreased in PSP group, PSP + Tan Ⅱ A group and PSP + Tan Ⅱ A + ADM group than in the control group (P 〈 0.05 ). The growth of implanted tumor was significantly inhibited by the Chinese medicines and chemotherapeutic drugs( P 〈 0.05 ). Thymus index of PSP + Tan Ⅱ A group was significantly increased vs control group ( P 〈 0.05) ;And thymus index in all therapeutic groups was significantly higher than in ADM control (P 〈 0.05). Conclusion: PSP and Tan Ⅱ A have inhibitory effects on the in vivo propagation
