复方JH-07抗肿瘤作用的研究

摘  要:目的研究复方JH-07体外和体内抗肿瘤活性作用,筛选抗肿瘤新复方。方法体外采用噻唑蓝(MTT)法检测JH-07对体外培养的人食管癌细胞EC9706、人白血病细胞K562耐药株及敏感株,人成纤维瘤细胞L929,人脑胶质瘤细胞TJ-905的生长抑制作用,计算生长抑制率及半数抑制浓度(IC50)。体内JH-07对H22肝癌荷瘤小鼠实体瘤的抗瘤实验采用常规抗肿瘤实验方法,用昆明种小鼠,雌雄各半,设立空白对照组、顺铂(0.005g·kg-1)及3个剂量JH-07给药组,每组10只小鼠;比较各组瘤重、肝、脾、胸腺指数,计算抑瘤率。结果JH-07可以显著抑制人白血病细胞K562耐药株及敏感株,人脑胶质瘤细胞TJ-905的生长,IC50分别为9.55,15.40,15.02μg·mL-1;JH-07小、中、高剂量组的肿瘤抑制率分别为48.7%,36.5%,46.7%。与对照组比较,均P<0.01。脏器指数及重量与空白对照组比较差异均无显著性(P>0.05),而与顺铂组比较差异有极显著性(P<0.01)。结论JH-07对体外培养的肿瘤细胞有较强的细胞毒性作用,可显著抑制细胞生长;体内可以明显抑制实体瘤生长。JH-07是值得深入研究的抗癌复方新药。Objective To study the anti-tumor effect of JH-07 in vitro and in vivo and identify a high-efficient and lowtoxic compound recipe. Methods Half-inhibitory concentration (IC50) was assessed in human esophageal cell EC9706, human leukemia cell K562 resistant line and sensitive line, human fibroblastic cell L929 and human glioma cell TJ-905 by MTT method. Meantime , mice model( n =40,10 mice in each group) transplanted liver cancer H22 were treated by JH-07 and DDP and tumor inhibitoryrate, thymus gland and spleen indexes, and liver weight were measured. Results IC50 of JH-07 in K562 resistant line, K562 sensitive line and TJ-905 were 9.55,15.40,15.02 μg · mL^-1, respectively. Tumor inhibitory rates of low, midest and high dose group in H22 mice were 48.7% ,36.5% and 46.7% ,and significantly higher than that of control group (P 〈0.01 ). However, JH-07 groups showed small differences in spleen index, thymus index and liver weight compared with control group (P 〉 0. 05 ). Nevertheless, compared with cisplatinum group, the differences were marked (P 〈 0. 01 ). Conclusion JH-07 had obvious anti-tumor activities in vitro and in vlvo, further study on JH-07 might be required.
