摘  要:目的研究液态冷藏条件(4±1℃)下,海藻糖保护剂对谷丙转氨酶(ALT)、谷草转氨酶(AST)、Y-谷氨酰转肽酶(GGT)、血清乳酸脱氢酶(LDH)、α-L-岩藻糖苷酶(AFU)等酶活性及其他生化指标的保护作用,并探讨海藻糖对酶等的最佳保护浓度,以提供一种人血清基质生化质控品的良好稳定剂。方法用惰性分离胶管(SSTII管)收集混合血清,加入保护剂;采用酶动力学方法分别在当天、第1、2、3、4、5、6、7、8、9、10个月分别测定5种酶活力及其他生化指标,并计算酶活力及其他生化指标保护率(%)。结果在10个月时,0.20mol/L海藻糖保护组的11种血清酶的平均保护率为92.6%,而无海藻糖保护组的11种血清酶的平均保护率为69.7%,海藻糖组的保护是无海藻糖组的1.32倍,海藻糖对酶等的最佳保护浓度为0.20mol/L。结论通过实验研究证实了海藻糖对人血清基质生化质控品液态冷藏状态下具有保护作用,是一种多酶等复合质控品的良好稳定剂。Objective To study the protective effects of a-Trehalose on the activity and the stability of enzyme at liquid refrigeration state (4±1 ℃ ), including serum ,ALT, AST, GGT, LDH and AFU and to explore the optimum protective concentration of a-Trehalose for the enzymes in order to provide a reliable quality control serum. Methods Mixed serum was collected in Insert Separation Gel Vacuum Collective Tube and in which the protective agent was added. The method of enzymatic kinetics was used to measure the activity of the five enzymes and other biochemical indexes on the same day and on the first, second, third, fourth, fifth, sixth, seventh, eighth, ninth and tenth month, respectively. Then the protective rate of enzymatic activity and other biochemical indexes were calculated. Result In the tenth month, the average protective rates of the eleven enzymes were 92.60% in the 0.20mol/L Trehalose group and 69.71% in the non-Trehalcse group, respectively. Trehalose group is 1.32 times as large as nou-Trehalose group. The optimum protective concentration of Trehalcse for the enzymes was 0.20mol/L. Conclusion The Trehalose has protective effect on enzymes at liquid refrigeration state and is a good stabilizer of multi-enzymes quality control material.
