摘 要:以甘蓝型油菜品种浙双758、浙双6号为材料,研究紫外线(UV)、甲基磺酸乙酯(EMS)和叠氮化钠(NaN3)对子叶外植体植株再生的影响。结果表明,UV处理子叶外植体后对植株再生有促进作用,当诱变时间达120和60 s时,两品种的愈伤组织诱导率和植株再生率分别达到最高值,显著高于对照。EMS不同浓度对长芽外植体数和植株再生数均有显著影响,且EMS处理浓度和处理时间存在互作效应。当EMS浓度达20mg/L,处理时间达6 h时,长芽外植体数和植株再生数达到最高。用浓度为500μmol/L NaN3,诱变时间为5,24h时处理子叶外植体所得到的再生植株数与浓度为100μmol/L,诱变时间为5,24 h所得到的结果差异显著,各处理因素间没有明显的互作效应。Effects of ultraviolet (UV) irradiation, mutagenic agents ethylmethane sulphonate (EMS) and sodium azide (NaN3 ) on plant regeneration of cotyledons in two Brassica napus cultivars (Zheshuang 758 and Zheshuang 6) in vitro were investigated. Resu]ts showed that UV had some promoting effects on cotyledon regenerations. The highest rates of callus induction and plant regeneration were observed fi'om the two cultivars when treated with UV for 120 s and 60 s, respectively. EMS at different concentrations significantly affected the numbers of shoot explants and plant regeneration, and there was interactive effect between EMS concentrations and treatment times. When EMS concentration was 20 mg/L and treatment time was 6 h, the highest numbers of shoot explants and plant regeneration were reached. There were sigsfificant differences in numbers of regenerated plants between different treatment eoneentrations of NaN3 (500 μmol/L or 100/μmol/L) and between different treatment times (5 h or 24 h). No obvious interactive effect was fotmd among different treatment factors.
