摘 要:目的研究亚低温对侧方液压冲击脑损伤大鼠海马MBP mRNA的早期表达变化的影响.方法雄性SD大鼠18只,随机分为3组:对照组(n=6);液压冲击脑损伤组(n=6),亚低温组(n=6).建立侧方液压冲击脑损伤大鼠模型,3 h后通过Real-time PCR法检测海马MBP mRNA变化.结果侧方液压打击脑损伤组与对照组比较,MBP mRNA相对表达量明显降低(P<0.01);亚低温组与侧方液压打击脑损伤组比较,MBPmRNA相对表达量明显增高,差异有统计学意义(P<0.01);亚低温组与对照组比较无统计学意义(P<0.05).结论亚低温可能通过增加MBP的表达减轻髓鞘降解及脱髓鞘作用来起到神经保护作用.Objective To study the early expression of MBP mRNA in the hippocampus of the lateral fluid percussion injury rats by randomization. Methods 18 male Sprague-Dawley rats were randomly divided into 3 groups: the control group (n=6) ; the fluid percussion injury group (n=6) , and the mild hypothermia group (n=6). We established the lateral fluid percussion animal models, and detected the change of MBP mRNA level in hippocampus 3 h after injury by Real-time PCR. Results Compared with the control group, the quantity of expression of MBP mRNA decreased obviously in the lateral fluid percussion injury group (P〈0:01). Compared with the lateral fluid percussion injury group, the quantity of expression of MBP mRNA increased obviously in the mild hypothermia group (P〈0.01). There was no difference in MBP mRNA level between the mild hypothermia group and the control group (P 〉0.05). Conclusion Mild hypothermia may act as neuroprotection by increasing the expression of MBP and easing the degradation of myelin sheath and demyelination.
