摘 要:在甘蔗外植体再生体系的基础上,在甘蔗愈伤组织继代、胚性愈伤组织芽分化以及幼芽生根阶段的培养基中附加甘露糖,研究甘露糖对甘蔗外植体再生的影响,以确定用于甘蔗遗传转化筛选的最适浓度.结果表明,甘露糖对甘蔗茎尖愈伤组织诱导、愈伤分化和幼芽根形成均产生抑制作用.愈伤组织生长、分化、生根3个阶段的抗性筛选浓度分别为3-5、4-5和3 g.L-1.The study was based on the regeneration system of sugarcane established by Key Lab of Eco-physiology & Genetic Improvement for Sugarcane, Ministry of Agriculture. Mannose of different concentrations was added into each medium of sugarcane tissue culture during secondary, regeneration and rootage stages. Moreover, the effect of mannose on explants regeneration was ana- lyzed, so that the optimum screening mass-concentrations needed by genetic transformation of sugarcane could be determined. The results indicated that mannose had inhibiting effect on induction frequencies of calli, shoots and rooting. The screening concentrations of mannose at the subculture, regeneration and rootage stages of sugarcane callus were 3 -5.4 -5 and 3 g· L^-1 . respectively.
