摘 要:目的:确定人参皂苷溶血测定中最优的实验条件。方法:用紫外分光光度法测定溶血度,分别对比了离心速度和时间、水浴时间、灭菌和非灭菌缓冲盐溶液及生理盐水溶液、不同酸碱度环境下的检测波长对实验结果的影响。结果:确定最优的实验条件为:用生理盐水做溶剂;溶血测定中溶液的水浴条件为37℃水浴2h;离心条件为转速3000r/min离心10min;在575nm处用紫外分光光度计测定其吸光度,并计算溶血度。结论:该方法稳定性和重现性均较好,适用于人参皂苷的溶血测定。Objective: To find out the optimal experimental conditions for hemolysis test of ginsenosides. Methods : Hemolytic degree was determined by eolorimetry, experimental conditions such as the effect of different centrifugal velocity and time, incubating time, sterilized,non-sterilized buffer solution and saline solution, detective wavelength in different pH value of solution were examined respectively.Results : The optimal conditions were determined to be:saline solution was adopted as solvent,the reaction mixture was incubated for 2h at 37℃, then, the mixture was centrifuged at 3.000r/min for lOmin and the absorbance of supernatant was determined at 575nm was measured. Conclusion : The method established showed good reproducibility and stability and can be used for the hymolysis evaluation of ginsenosides.
