摘  要:目的探讨曲古押菌素A(trichostatin A,TSA)对甲状腺鳞癌细胞株SW579生长增殖的影响。方法分别以终浓度为25、50、100、200、400 nmoL/L的TSA作用于SW579细胞株,在倒置显微镜下观察细胞生长情况及形态变化;采用四甲基偶氮唑蓝比色法(MTT)检测细胞的体外增殖、吖啶橙染色荧光显微镜观察细胞形态及细胞凋亡、流式细胞术检测细胞凋亡率的改变。结果TSA作用后对细胞的增殖有抑制作用;吖啶橙染色后光镜下观察发现有典型的凋亡小体生成;流式细胞仪分析凋亡率随TSA浓度的升高而升高(P<0.05)。结论不同浓度的TSA可抑制SW579细胞增殖,促进细胞凋亡,且呈剂量依赖性。Objective To study the effects of trichostatin A (TSA) on apoptosis and proliferation of thyroid squamous cell carcinoma SW579. Methods Treated with different doses of TSA (final concentration of 25 nmol/L, 50 nmol/L, 100 nmol/L, 200 nmol/L, 400nmol/L), the cells were observed with an inverted microscope; The proliferation activities of cell line were determined by using methyl thiazolyl tetrazolium (MTT) assay, Cell morphologies and apoptotic were observed by fluorescence microscope stained with acridine orange , Flow cytometry was adopted to examine apoptotic rate. Results TSA inhibited significantly the growth of cell line and the effect was with a dose dependent manner. Typical apoptotic morphologic feature was discovered under fluorescence micro- scope. The apoptotic peak was detected with FCM and apoptotic rate increased with the concentration raised, Conclusions TSA can inhibit the proliferation and induce apoptosis in the thyroid cancer cells with a dose - dependent manner.
