摘 要:为明确小麦红叶耳基因的染色体位置,以小麦BS210(红叶耳)/O201(绿叶耳)的DH群体为材料,于2005~2006和2006~2007年度分别将120个DH株系及亲本在北京种植,并对该群体进行叶耳颜色的鉴定。遗传和方差分析表明,红叶耳由一对主效基因控制,同时受环境影响,并存在基因与环境互作效应;根据DH群体叶耳颜色分别构建红叶耳池和绿叶耳池,利用BSA法和SSR引物筛选与红叶耳基因连锁的标记,将红叶耳相关基因定位于3B染色体长臂上,QTL分析表明,红叶耳主效QTL位于WMC182和WMC54之间,贡献率为24.1%,与最近标记WMC182的距离为7.1cM,红叶耳基因来自母本BS210。本研究结果还表明,BSA与QTL方法相结合,有利于提高简单性状基因定位的效率和精确度。Red-auricle is one of important agronomic characters in wheat. It is easily influenced by en- vironment. Studies on genetic mapping of red-auricle using molecular markers were few reported in wheat. Identification of red-auricle genes and their closely linked molecular markers is very important for developing wheat cultivars. One DH population with 120 lines derived from a cross between BS210 and O201 was sown with two replicates in Beijing. The color investigation of auricle was carried out before tasseling during 2005-2006 and 2006-2007 cropping seasons. With the analysis of variance, the result showed that red-auricle is controlled by one pair of major gene, and it is influenced by he- redity, environment and exists interaction between gene and environment. The major red-auricle gene was mapped to chromosome 3B using Simple Sequence Repeat (SSR) markers and bulked segregant analysis (BSA). Partial linkage maps around red-auricle loci in chromosomes 3B was constructed using the 120 lines of the DH population. With quantitative trait location (QTL) analysis, the major red-au- ricle gene of wheat was detected between the marker WMC182 and WMC54 in the 3BL, accounting for 24.1% of phenotypic variance. The distance to the nearest marker WMC182 is 7.1 cM, and the red- auricle gene derived from female parents BS210. This study demonstrated the feasibility in gene map-ping using bulked segregant analysis coupled with quantitative trait location(QTL)for simple traits.Also the results of mapping and molecular markers on red-auricle gene would be useful to identify the traits for varieties of wheat.
