摘 要:目的通过建立变应性鼻炎(allergic rhinitis,AR)豚鼠模型,探讨嗜酸细胞活化趋化因子(eotaxin)对鼻黏膜中嗜酸细胞(eosinophils,Eos)局部浸润及活化的作用,进一步阐明Eos在鼻黏膜的摹集及其活化的机制。方法卵清蛋白致敏法制备AR豚鼠模型,分别采用免疫组化、逆转录聚合酶链反应技术检测eotaxin蛋白及mRNA在鼻黏膜中的表达,免疫荧光法测定鼻腔冲洗液中嗜酸细胞阳离子蛋白(eosinophil cationic protein,ECP)含量标示Eos活化程度,鼻黏膜HE切片每高倍视野下Eos平均个数标示Eos浸润程度。结果与对照组相比,AR模型组鼻黏膜中eotaxin蛋白阳性表达率、eotaxin mRNA表达、Eos个数以及鼻腔灌洗液中ECP含量均显著增加(P<0.001,P=0.006,P<0.001,P<0.001);相关分析表明eotaxin表达与Eos浸润程度及ECP含量呈正相关(r分别=0.849、0.691、0.656、0.819)。地塞米松干预后以上指标一致性下降。结论AR豚鼠鼻黏膜中eotaxin表达增强,是Eos局部浸润和活化的重要影响因素;地塞米松可以通过降低AR豚鼠鼻黏膜中eotaxin的表达减少鼻黏膜中Eos的局部浸润和活化。OBJECTIVE To investigate the effect of chemotatic factor (eotaxin) on local infiltration and activation of eosinophiles through establishing allergic rhinitis animal model, and explore the mechanism of eosinophiles infiltration and activation in nasal mucosa further, METHODS The allergic rhinitis animal model was established by being challenged with ovalbumin. Protein and mRNA expression of eotaxin was detected by immunohistochemical method and RT-PCR, and content of eosinophil cationic protein (ECP) in the nasal cavity lavaging fluid (NLF) was detected by immumofluorescence method. They were considered as the extent of eosinophiles activation. Average eosinophiles number of per high power field in nasal mucosa HE section was considered as infiltrative extent of eosinophiles. RESULTS In the allergic rhinitis group without treated, positive rate and mRNA expression of eotaxin in nasal mucosa, content of ECP in NLF and eosinophiles number in nasal mucosa were all increased compared with those in normal control group (P〈0.001, P=0.006, P〈 0.001, P〈 0.001). After dexamethasone treated, they were all decreased compared with those in allergic rhinitis group. Associative analyses indicated that there was a notable right relation between eotaxin and ECP, eosinophile (r=0.849, 0.691, 0.656, 0.819) . CONCLUSION Expression of eotaxin in allergic rhinitis guinea pigs nasal mucosa is enhanced highly, which is an important factor in local infiltration and activation of eosinophiles. Dexamethasone can decrease the infiltration and activation of eosinophiles in nasal mucosa through reducing the expression of eotaxin.
