大鼠肝叶切除后肝细胞再生相关蛋白的蛋白质组学分析

摘  要:目的通过对肝叶切除后6、24 h的肝细胞与正常肝细胞的蛋白质组分析,筛选出与肝细胞再生相关的蛋白质。方法选择雄性SD大鼠9只(体质量200 g),分3组,6 h组、24 h组和对照组,每组3只,建立大鼠70%肝叶切除模型。6 h组在肝叶切除后6 h分离肝细胞,24 h组在肝叶切除后24 h分离肝细胞,对照组为正常肝细胞。裂解液裂解细胞提取总蛋白,运用蛋白组学相关技术分析3组细胞的蛋白组学差异。结果总共得到47个差异蛋白质点,鉴定出42个。结论我们的数据显示:24 h组与对照组和6 h组都有重叠,而6 h组和对照组重叠部分较少,推测其可能的原因为6 h组肝细胞处于肝再生的启动阶段,与对照组差异较大,而24 h组,肝再生已启动,各种维持肝细胞正常功能的蛋白质也开始表达。Objective To screen proteins related with liver regeneration by studying the proteomic difference between the hepatocytes 6 h and 24 h after the 70% hepatotectmy and the normal ones. Methods Nine male SD rats (weight 200 g) were selected and randomly divided into 3 groups: the control group, the 6 h group and the 24 h group, each one with 3 rats. The rats 70% hepatectomy model was built. The hepatocytes in the control group were immediately isolated from the rat liver and those in the 6 h group and 24 h group were isolated after 6 h and 24 h respectively. We extracted the total proteins with cell lysis and anselyzed the proteomic differences between the 3 groups. Results We found 47 different proteins and identified 42 of them. Conclusion The results indicated that the proteins produced in the 24 h group are to a large extent identical with those in the 6 h group and the control group, however, the proteins produced in the 6 h group are seldom the same with those in the control group. This is because the hepatocytes in the 6 h group was in the start state of liver regeneration while the hepatocytes in the 24 h not only produced proteins related to the liver regeneration but also the proteins maintaining the normal physiological function of the hepatocytes.
