摘 要:目的评价促进造血干细胞增殖与血红蛋白合成营养组合物对小鼠再生障碍性贫血造血功能的影响。方法BALB/c小鼠100只,适应性喂养1周后,随机分为:正常对照组、再障模型组及不同剂量的营养组合物组。采用乙酰苯肼、X射线、环磷酰胺联合应用的方法建立再生障碍性贫血小鼠模型,以铅砖屏蔽施以假照射及单纯等量生理盐水相应部位注射为正常对照组。试验第7天开始,营养组合物高、中、低剂量组小鼠每天灌胃,分别给予1445.55,963.7,674.59 mg/(kg·d)营养组合物,直至第45天,颈椎脱臼法处死小鼠,观察试验小鼠的血象、骨髓象、造血细胞内的线粒体、造血干细胞集落的形成及肝脏、脾脏的病理变化。结果营养组合物组的外周血象三系细胞、骨髓有核细胞数、造血干细胞集落的形成明显高于再障模型组(*p<0.05,*p<0.01),且呈剂量-效应关系,促红细胞生成素(EPO)因代偿作用含量显著降低(*p<0.01)。骨髓透射电镜显示,不同剂量的营养组合物组与再障模型组比较,同类造血细胞内线粒体数目明显增多(*p<0.01)。造血干细胞集落实验证明,营养组合物组集落形成率明显高于再障组。不同剂量的营养组合物组与再障模型组相比,肝脏组织水肿减轻,小叶结构明显清晰,肝细胞形态正常,脾脏虽仍有部分充血,但脾小体个数较再障模型组明显增多。结论营养组合物促进再障小鼠外周血细胞生成和骨髓造血干细胞的增殖,促进骨髓造血细胞内线粒体的增加,对肝、脾的损伤具有明显恢复作用。Objective To evaluate the function of the nutrients mixture to promote hematopoietic stem cell proliferation and hemoglobin synthesis in aplastic anemia (AA) mouse. Method 100 BALB/c type mice were fed and divided randomly into three groups: normal control, AA model and the different doses of the nutrients mixture groups. The AA mouse model was established by combination of hydracetin, X-ray and cyclophosphamide, the normal control by lead shielding irradiation and the saline injection. After the seventh day, mice were fed by nutrients mixture in a dose-dependent manner (1445.55, 963.7, 674.59 mg / kg d). The mice were killed by cervical dislocation ,and then blood, bone marrow, mitochondria of hematopoietic cells, hematopoietic stem cell colony formation and liver, spleen pathological changes were observed 45 clays later. Results The blood cells, nucleated cells of bone marrow, hematopoietic stem cell colony formation in nutrients mixture group were significantly more than those in AA model (*p〈0.05, *p 〈0.01), with a dose dependent. The erythropoietin (EPO) significant reduced by compensatory role (*p〈0.01), the number of mitochondria in similar cells markedly increased (*p〈0.01) ,the colony formation rates markedly increased (*p〈0.01) in nutrients mixture groups than in AA group. The liver tissue edema was reduced and lobular structure was clear ,the number of splenic body was also increased in nutrients mixture groups than in AA group. Conclusion Nutrients mixture improves the AA mice bone marrow damage, promotes blood cell production and hematopoietic stem cells proliferation, as well as increases the number of mitochondrial and recovers damaged liver and spleen of AA mice.
