摘　　要：目的探讨曲伐沙星抗炎作用的分子机制。方法将分离的单核细胞预先用TVF孵育2h,随后用LPS刺激,ELISA检测TNF-α和IL-6的诱生情况,并用RT-PCR检测其mRNA的表达;提取核蛋白,Western blot分析NF-κB激活情况。结果TVF能以剂量依赖方式抑制单核细胞产生TNF-α和IL-6,并抑制其mRNA的表达;同时TVF能抑制NF-κB的激活。结论TVF能抑制单核细胞产生TNF-α和IL-6,这可能是其抗炎症作用的方式之一,该效应可能与其抑制NF-κB的激活有关。Objective To investigate the anti-inflammatory activity of trovafloxaein （TVF）. Methods Isolated monocytes were preincubated with TVF for 2h, and then stimulated by LPS. Induction of TNF-α and IL-6 was detected by ELISA, and their mRNA levels were assessed by RT-PCR; Nucleoprotein were extracted and activation of NF-κB was detected by Western blot. Results TVF inhibited monocytes production of TNF-α and IL-6 in a dose-dependent manner, and inhibited their mRNA expression, NF-κB was also inhibited by TVF either. Conclusion The anti-inflammatory activity of TVF occurrs by inhibit TNF-α and H.-6 production, this effect is probablely mediated by inhibited NF-κB activation.
