摘 要:[目的]为嗜盐菌HBCC-3的应用研究奠定基础。[方法]从连云港台南盐场分离出嗜盐菌HBCC-3,通过正交试验对其培养条件进行优化。通过PCR方法扩增HBCC-3的16SrDNA,对其进行同源性比较和系统发育学分析。[结果]嗜盐菌HBCC-3为乳白色,杆状,革兰氏染色呈阳性,菌落大小为4~6 mm。温度对HBCC-3生长影响最大,其次是摇床转速、NaCl、柠檬酸三钠、MgSO4.7H2O、pH值,而KCl最小。HBCC-3的最适培养条件为:温度35℃、pH值9.0、摇床转速200 r/min、柠檬酸三钠2.0g/L、NaCl 10%、KCl 1.0g/L、MgSO4.7H2O 10.0 g/L。菌株HBCC-3与碱杆菌属菌株Alkalibacillus salilacus BH165的相似性最高,达99%。[结论]菌株HBCC-3与Alkalibacillus salilacus[Objective] The aim of the research was to lay the foundation for the application research of halophilic bacteria HBCC-3.[Method] Halophilic bacteria HBCC-3 was isolated from Tainan salt field of Lianyungang and its culture conditions were optimized in the orthogonal test.16S rDNA of HBCC-3 was amplified by using PCR method to make the homology comparison and the phylogenetic analysis.[Result] Halophilic bacteria HBCC-3 was ivory white,being rod-shaped,showing Gram-positive,with the size of bacteria colony being 4-6 mm.The effect of temperature on the growth of HBCC-3 was greatest,followed by shaker rotating speed,NaCl,trisodium citrate,MgSO4·7H2O and pH value,and that of KCl was smallest.The optimum culture conditions of HBCC-3 were as follows: the temperature of 35℃,pH value of 9.0,the shaker rotating speed of 200 r/min,trisodium citrate 2.0g/L,NaCl 10%,KCl 1.0 g/L,MgSO4·7H2O 10.0 g/L.The similarity between Strain HBCC-3 and Strain Alkalibacillus salilacus BH165 of Alkalibacillus was highest,reaching 99%.[Conclusion] Strain HBCC-3 and Alkalibacillus salilacus BH165 was the same species.
