新疆南疆地区棉花枯萎病菌的致病力测定与遗传多样性分析

摘　　要：[目的]明确新疆南疆地区棉花枯萎病菌致病性变异和遗传多样性,为南疆地区棉花抗病育种和棉花枯萎病的综合防治提供依据。[方法]选用12个棉花枯萎病菌(Fusariumoxysporum)代表菌株在4个长绒棉品种上进行致病力测定和AFLP分析。[结果]根据病情指数将南疆不同团场的菌系致病力划分为中、弱2个类群;方差分析和对应分析的结果表明,不同菌系之间的致病力存在极显著差异,4个海岛棉品种和12个菌系可聚集成4类。选用9对EcoR I和Mse I引物组合,对供试菌株进行遗传多样性研究,结果表明,在0.52的相似水平上,全部菌株被分为4个AFLP群。[结论]新疆南疆地区不同团场间的棉花枯萎病菌存在明显的致病性分化,显示出较大的遗传多样性。[Objective] The research aimed to make clear the pathogenicity variation and genetics diversity of Fusarium oxysporum f.sp.vasinfectum in Southen Regions of Xinjing,and provide the basis for disease-resistant breeding and integrated control of cotton wilt.[Method] Twelve strains of Fusarium oxysporum f.sp.vasinfectum isolated from the Long-libre Cotton regions of Xinjing were tested based on the symptom and pathogenicity in the cotton cultivars and analyzed genetic diversity by using AFLP technology.[Result] According to the average disease index on four differential Long-libre Cotton cultivars,the stains were differentiated into 2 groups.The results of variance analysis and corresponding analysis of the average disease index of 12 tested strains on 4 differential Long-libre Cotton cultivars showed that the pathogenicity in different strains was very significant difference and were divided into 4 groups.Twelve strains were analyzed by AFLP fingerprinting.The result showed that all tested strains were divided into 4 AFLP groups at the similarity level of 0.52.[Conclusion] The study showed that there existed obvious differentiation among the pathogenicity of Fusarium oxysporum f.sp.vasinfectum in Southen Regions of Xinjing,and AFLP groups revealed a great genotypic diversity.
