羊Ⅰ型前胶原的提取、纯化与抗血清的制备应用于放免法的建立

摘　　要：目的建立测定血清Ⅰ型前胶原(procollagen typeⅠ,PCⅠ)放射免疫分析法(放免法),应用于前列腺癌骨转移的诊断。方法从羊胎皮中提取PCⅠ免疫兔获取特异性抗血清。结果经聚丙烯酰胺凝胶电泳证实为单一区带,β疏基乙醇还原后,出现I型前胶原特有的链。用此抗原免疫兔获得高效价抗血清,效价为1∶128倍和1∶64倍以上。结论抗原、抗体可用于PCⅠ放免法的建立。Objective To establish the radioimmunoassay（RIA） method for the measurement of type Ⅰ procollagen（PCⅠ） and diagnosing the bone metastasis of prostate cancer. Methods PC Ⅰ was extracted from the sheep fetus skin and the specific antiserum was obtained from the immune rabbit. Results The isolated zone was certified by PAGE. There were the specific chains after reduction of β-mercaptopurine. From the immune rabbits,the high titer antiserum was obtained. The titer was over 1 ： 128 and 1 ： 64. Conclusion Antibody and antigen can be used in the establishment of type Ⅰ procollagen radioimmunoassay.
