摘 要:目的:探讨银杏叶提取物(EGb)对哮喘大鼠气道上皮杯状细胞增生和黏液高分泌的抑制作用,以及对哮喘大鼠血清及肺泡灌洗液中IL-13表达水平的影响。方法:以卵蛋白激发制作大鼠哮喘模型,EGb组腹腔内注入EGb作为干预因素,分别以HE、阿辛兰-PAS染色观察气道上皮的改变,杯状细胞的表达及黏液生成,ELISA法检测血清及支气管肺泡灌洗液(BALF)白细胞介素13(IL-13)浓度。结果:哮喘组较对照组气道上皮杯状细胞明显增多,黏液生成增多,血清及肺泡灌洗液中IL-13表达水平显著增高。EGb组较哮喘组杯状细胞数量显著减少,黏液生成减低,血清及肺泡灌洗液中IL-13表达水平明显降低。结论:EGb可以通过抑制IL-13的表达水平减轻哮喘大鼠气道黏液高分泌。Objective: To observe the effects of the extract of Ginkgo biloba(EGb) on mucus hypersecretion of asthmatic rats and the expression of IL-13 in serum and bonchoalveolar lavage fluid(BALF), Method: Rats were randomly divided into 3 groups :normal rats ,asthmatic rats , EGb treated rats. Asthmatic rats were sensitized with ovalbumin(OVA). The assessment of IL-13 in both serum and BALF was performed, histochemical analysis and Alcian blue- periodic acid Schiff (PAS) staining to observe goblet cell. Result: Alcian blue-PAS staining revealed more mucus and goblet dells in the asthmatic bronchial epithelial compared to control rats . meanwhile EGb obviously suppressed the production of mucus and hyperplasia of global cells of asthmatic rats . The productions of IL-13 both in serum or in BALF in asthmatic rats were increased significantly from the normal rats , EGb treatment could strongly reduce the production of IL-13 in both serum and BALF. Conclusion: EGb significantly suppressed the mucus hypersecretion and global cells hyperplasia in asthmatic rats by inhibiting the production of IL- 13.
