摘  要:目的观察三七总皂甙(PNS)对脑缺血再灌注损伤大鼠细胞间粘附分子1(ICAM-1)表达的影响。方法采用线栓法制备大鼠大脑中动脉阻断局灶性脑缺血模型,于脑缺血再灌注24h后断头取脑,用免疫组织化学法检测脑组织中ICAM-1表达,苏木精-伊红染色对脑组织进行病理学检查,光镜下观察脑血管周围白细胞的粘附与渗出。结果PNS能明显抑制大脑皮质及尾壳核ICAM-1的表达,减轻白细胞的粘附与浸润。结论PNS对脑缺血再灌注后的炎性损伤的保护作用可能与抑制ICAM-1表达,减轻白细胞的粘附与浸润有关。Objective: To study effect of Panax Notoginseng Saponinsa(PNS) on the expression of intercellular adhesion molecule-1(ICAM-1) in Rats with Cerebral Ischemia Reperfusion Injury. Methods: The model of local cerebral ischemia-reperfusion was prepared by the intravascular thread occlusion of the middle cerebral arteries(MCAO). The middle cerebral arteries of rats were occluded for 2 hours and reperfused for 24 hours. The expression of ICAM-1 was observed by immunohistochemical staining. The adhesiveness and infiltration of leucocytes were observed by HE staining. Results: PNS was shown to markedly inhibit the expression of ICAM-1, alleviated the adhesiveness and infiltration of leucocytes. Conclusion: PNShas protective effects on inflammatory injury following Cerebral Ischemia Reperfusion Injury in Rat by inhibiting ICAM-1 expression, alleviating the adhesiveness and infiltration of leucocytes.
