摘 要:[目的]了解大连地区耐环丙沙星的大肠埃希菌中qnrA基因的流行情况并研究其耐药机制。[方法]收集临床分离的耐环丙沙星的大肠埃希菌213株,通过PCR法扩增qnrA基因,对qnrA基因阳性株,KB纸片法检测抗菌药的体外抗菌活性,琼脂稀释法测MIC,并进行质粒结合试验。[结果]213株菌中查到2株qnrA阳性菌株,测序结果与Genbank中qnrA基因比对吻合率达100%。其中一株对多种抗菌药耐药,另一株则比较敏感。[结论]大连地区检测到了qnrA基因阳性的菌株,耐药机制复杂,应引起临床重视。[ Objectives ] To investigate the prevalence of qnrA gene in Escherichia coli in Dalian, and study it' s mechanism of the resistance. [ Methods] A total of 213 clinical isolates of quinolone resistance in Escherichia coli. The qnrA genes were amplified by PCR. Kirby - Bauer disk diffusion method was used to test the in vitro susceptibility of the isolates to anti- microbial agents. MIC values were determind for qnrA gene positive strains. Plasmid conjugation experments were done in order to investigate the qnrA genes can transfer or not. [ Results ] The qnrA genes were identified in 2 clinical isolates and the qnrA gene sequence matched that of GenBank with 100%. One was resistant to many antimicrobial agents,another was not. The plasmid conjugation tests were carried out in the both strains. [ Conclusions] The qnrA genes have been existing in DaLian. The serious situation of complex resistance mechanism should be highly concerned in clinics.
