次氮三乙酸铁对大鼠糖代谢影响的研究

摘  要:目的了解体内铁超负荷对大鼠糖代谢的影响。方法雄性Wistar大鼠随机分四组:铁干预组(A组)、次氮三乙酸二钠对照组(B组)、铁加去铁干预组(C组)、空白对照组(D组)。观察腹腔葡萄糖耐量试验(IPGTT),测定血清铁(SI)、血清铁蛋白(SF)、血清丙二醛(MDA)变化。取胰腺作铁染色行积分光密度(IOD)值分析。结果(1)A、C组大鼠体重增长明显低于B、D组(p值<0.05)。(2)A、C组SI、SF、MDA、胰腺铁染色IOD值显著高于B、D组(p值<0.05)。(3)A、C组胰腺组织可见普鲁士蓝颗粒沉积。(4)第10周时A、C组基础血糖及2小时血糖较B、D组高(p=0.059,p=0.043),A组2小时胰岛素水平有降低趋势;干预后A、C组基础血糖及2小时血糖较干预前高(p<0.05)。结论(1)给Wistar大鼠反复腹腔注射FeNTA八周后建立铁超负荷大鼠模型。(2)铁超负荷大鼠胰腺组织可见铁沉积。(3)铁超负荷大鼠脂质过氧化产物明显增高。(4)铁超负荷大鼠糖耐量受损,β细胞分泌胰岛素能力有降低趋势。Objective To explore the effect of iron overload on glycometabolism of the Wistar rats. Methods Wistar male rats were divided randomly into four groups. Group A received FeNTA. Group B received Na2NTA, Group C received DETAPAC and FeNTA . Group D was untreated control. Intraperitoneal glucose tolerance tests (IPGTT) were performed. Serum iron (SI) , serum ferritin (SF) , and serum malondialdehyde (MDA) were measured. Portions of pancreas of each rat were collected for Prussian blue stain. The values of IOD of iron granules of pancreas were analyzed . Results ( 1 ) Group A and C showed a lower growth rate than group B and D (p 〈0. 05). (2) The levels of SI,SF,MDA,IOD of iron granules in group A and C were higher than group B and D (p 〈0. 05). (3) Prussian blue positive granules were observed in parenchymal cells of pancreas in group A and C. (4) At lOth week, the basal plasma glucose (BPG) and plasma glucose 2 hours after the glucose load (2hPG) of group A and C were higher than group B and D(p = 0. 059, p = 0. 043 ) , and the plasma insulin of group A had a tendency to decrease ; Group A and C showed elevated levels of BPG and 2hPG after intervention (p 〈 0. 05 ). Conclusions ( 1 ) An iron overload rat model was developed by repeated injections( i. p. ) of FeNTA for 8 weeks. (2) Iron granules were found to deposit in pancreas of the model. ( 3 ) Iron overload rats had higher level of lipid peroxidation in vivo. (4) Iron overload in vivo impaired glucose tolerlance and capacity of β cells secrete insulin.
