摘 要:水泡性口炎病毒(VSV)经80℃2小时和100℃30分钟干热灭活后,其细胞感染试验与PCR分析结果并不一致。细胞感染试验为阴性,但用PCR方法却仍能检出VSV的核衣壳基因片段。这种结果提示,在一定条件下,病毒核酸的热变性可能不是断裂,或断点不在扩增片段范围之内。故用PCR方法评价热力灭活病毒效果时应慎重。n order to know the difference of evaluation of inactivation efficacy by cell infection method and PCR method in virus inactivation test, test for inactivation of vesicular stomatitis virus (VSV) by heat was used for observation and comparison of these 2 methods.The results indicated that after dry heat inactivation of VSV at 80℃ for 2 hr and 100℃ for 30 min, the cell infection test was negative, while PCR could still detect fragments of nuclear capsid gene. When dosage of heat was increased to 80℃ for 72 hr or 100℃ for 2 hr, PCR test was still positive. This indicates that under certain conditions, heat denaturalization of viral RNA may be not fragmentation or the break site not located in amplification fragment, therefore it should be careful to use PCR method for evaluating the efficacy of heat in inactivating viruses.
