摘 要:目的探讨肝细胞生长因子(hepatocyte growth factor,HGF)对周围神经损伤修复的作用。方法以Wistar大鼠坐骨神经挤挫伤为动物模型,神经损伤局部肌肉注射腺病毒介导的人肝细胞生长因子(Ad-HGF),与无注射HGF组对照,通过步态分析、肌肉湿重测定、计算机图像分析等指标评价神经损伤后再生效果。结果术后4周,在坐骨神经指数、肌肉湿重恢复、计算机图像分析等再生指标的比较中,注射HGF组明显强于对照组(P〈0.05)。结论HGF能促进大鼠坐骨神经损伤后的有髓纤维再生和功能恢复,是一种有效的治疗周围神经损伤的神经营养因子。Objective To explore the effect of hepatocyte growth factor on peripheral nerve regeneration. Methods Sciatic nerve contusion injury was made by a custom-made clamp in Wistar rats, in which human hepatocyte growth factor expressed by adenoviral vector (Ad-HGF) was injected into the muscle around the injured nerve. The results of nerve regeneration were evaluated by sciatic nerve function index (SFI) , muscle wet weight, neural electrophysiology and image analysis. Results Four weeks after sciatic nerve injury, the results of sciatic nerve function index (SFI) , muscle wet weight, neural electrophysiology and image analysis showed better nerve regeneration in group injected with HGF than control group (P 〈 0.05 ). Conclusion Hepatocyte growth factor can promote axon regeneration and functional recovery and is an effective neurotrophic factor for peripheral nerve regeneration after injury.
