泥蚶线粒体COI和16S rRNA基因片段序列研究

摘　　要：采用PCR技术扩增了泥蚶线粒体DNA的COI和16S rRNA基因片段,PCR产物经T载体连接后进行克隆、测序,用MEGA version 3.0软件计算这2个基因片段序列碱基组成.结果显示:COI和16S rRNA基因删除引物序列后分别得到660bp和548bp的核苷酸序列,T,C,A和G碱基含量分别为40.4%,14.7%,20.6%,24.3%和23.2%,25.9%,30.3%,20.6%.COI和16S rRNA基因片段对泥蚶不同地理种群的遗传分化的研究具有一定的应用价值.Mitochondrial cytochrome c oxidase subunit I gene（COI） and 16S rRNA gene fragments of Tegillarca granosa are amplified using PCR, then the PCR products are ligated into T-vectors, cloned and sequenced. 660 bp and 548 bp nucleotide sequences of partial COI gene and partial 16S rRNA gene are obtained respectively. Nucleotide composition frequencies are analyzed with the MEGA 3.0 software. The contents of T, C, A and G are found to be 40.4%, 14.7%, 20.6%, 24.3% in COI; 23.2%, 25.9%, 30.3%, 20.6% in 16S rRNA.
