摘  要:首次报道了在中国东海海区两个不同水样中获得的有毒裸甲藻赤潮种Karlodinium micrum的显微观察和分子鉴定。两株K.micrum纯系种质是对两次东海海区不同来源的水样(2005年6月东海洞头海区以米氏凯伦藻(Karenia mikimotoi Hansen)为优势的赤潮水和2006年4月东海渔山海水自然生态保护区的水样)进行选择演替和纯种分离发现的。报道了对两株藻进行光学显微镜和扫描电子显微镜观察结果,以及基于ITS基因测序的分子鉴定结果。显微观察发现,K.micrum为小型单细胞裸甲藻,细胞长略大于宽,平均长宽分别为7.1μm和5.7μm,横沟深裂,顶沟明显、直裂。细胞具有一条纵鞭毛和一条横鞭毛,细胞游动迅速。ITS基因测序发现,源于东海的两株K.micrum与GenBank中Karlodinium micrum的相似度为98~99%。系统发生分析显示,该两株K.micrum与GenBank中Karlodinium micrum归于一类,与其它裸甲藻距离较其与具甲板甲藻的距离近。In this paper, two pure cultures of toxic unarmoured dinoflagellates Karlodinium micrum from different sites in the Eastern China Sea (ECS) were first reported and identified with microscopic observations and molecular analysis. The two strains of K. micrum pure cultures in this paper were collected from two different sites in the ECS,including the site: from Dongtou red tide samples while Karenia mikimotoi was the dominant species in June 2005; and the site from Yushan natural sea water protection areas in the ECS in April 2006. Light microscope and SEM observations and ITS analysis were done on both of the strains. From the microscopic observations, K. micrum was small unicellular athecate dinoflagellate. Its length was slightly longer than its width. The average length and width were 7.1 μm and 5.7 μm respectively. The broad cingulum was deeply excavated. A well-defined apical groove was present ventrally on the anterior of the cell. One transverse flagellum and one longitudinal flagellum occurred. Cells were found swimming fast in the water. Molecular analysis by ITS assay showed that, two strains of K. micrum from the ECS showed 99% similarity with K. micrum from GenBank online, and they were classified into the same rank with K. micrum, very near to some other athecate dinoflagellates, further away from armoured dinoflagellates.
