骨髓来源成骨细胞体外成骨表达的研究

摘　　要：目的探索在体外诱导兔骨髓基质干细胞(bone marrow stem cells,BMSCs)向成骨细胞分化及成骨表达的特性。方法抽取兔骨髓,通过离心法获取单个核细胞,在体外经条件培养基培养21d,通过倒置显微镜及激光共聚焦显微镜观察成骨细胞的形态学特点;应用MTT法测定成骨细胞活性;并检测成骨细胞分泌碱性磷酸酶活性及形成矿化结节情况。结果体外BMSCs可在成骨条件培养液下诱导培养后可向成骨细胞分化,经倒置显微镜和激光共聚焦显微镜观察证实,诱导后BMSCs由长梭形转变为短胖形的成骨细胞,MTT检测成骨细胞活性良好,分泌碱性磷酸酶并形成矿化结节。结论骨髓基质干细胞在体外可定向诱导为成骨细胞,并具有成骨细胞功能,有希望成为理想的种子细胞应用于临床。Objective To study the bone tissue osteogenesis ability of osteoblast induced by bone marrow stem cells（BMSCs） in vitro. Methods The BMSCs were separated by gradient centrifugation on percoll from rabbit bone marrow, then observing and testing the biologic characters of BMSCs induced by culturing with a conditional medium for 21day. Results Osteoblast can be induced from BMSCs by conditional medium, moreover it can secret ALP and format the ealcifie nodules. Conclusion BMSCs can be induced successfully to osteoblast and express the osteogenesis function in vitro and can be used as ideal seed cells for bone tissue engineering.
