摘 要:目的观察极低密度脂蛋白受体(VLDLR)的总量及两种亚型在正常(NC)大鼠与2型糖尿病(T2DM)大鼠各种组织(心、脑、肾、肌肉、脂肪)中的分布与表达。方法取NC大鼠与T2DM大鼠的各组织,提取mRNA,用半定量PCR方法进行扩增。结果T2DM大鼠各组织中的VLDLR总量与NC组相比有不同程度下降,在脑、肾、肌肉、脂肪组织中下降明显(P<0.01)。在脑、肾脏组织中I型与II型VLDLR均有明显下降,肌肉组织以I型下降为主(P<0.001),脂肪组织以II型下降为主(P<0.001)。结论T2DM胰岛素抵抗与血脂紊乱状态影响了VLDLR的分布与表达。Objective To investigate the distribution and expression of total amount and two subtypes of very low density lipoprotein receptor(VLDLR) in various tissues (heart, cerebrum, kidney, muscle, adipose)of normal SD rats and type 2 diabetic SD rats. Methods The mRNA of VLDLR from various tissues were amplified by semi-quantitative RT-PCR. Results The expression levels of VLDLR were reduced in all collected tissues compared with normal ones, especially in cerebrum, kidney, muscle, adipose(P〈0.01). The type I and type II VLDLR expressions were all decreased in cerebrum and kidney. In muscle, type I VLDLR expression was sharply reduced (P〈0. 001), but in adipose, type II VLDLR was greatly decreased(P〈0. 001). Conclusions The state of insulin resistant and disorder of lipid metabolism in type 2 diabetes can affect the expression and distribution of VLDLR in tissues.
